Molecularly imprinted polymers (MIPs) are crosslinked polymeric materials specifically designed to exhibit high binding capacity and selectivity toward a target molecule (template), which is intentionally introduced during the synthesis process. In addition to growing interest in the various formats and applications of these polymers, considerable attention has been devoted to gaining a deeper understanding of the heterogeneous nature of their binding sites and to accurately quantifying template-MIP affinity. This lecture will illustrate how MIP characterization can benefit from the application of analytical separation techniques. In this context, and with specific reference to my own research, examples will be presented involving zonal liquid chromatography, frontal affinity chromatography (FAC), capillary electrochromatography (CEC), solid phase extraction (SPE) and affinity capillary electrophoresis (ACE). General principles, methodologies and experimental details will be discussed, to provide a clearer understanding on how these techniques operate and on their advantages or limitations.
